چکیده

Osteoporosis is a complex bone disease indicating porous with low mass density and fragility. Cathepsin K, V-ATPase, αVβ3 integrin are exhibited as novel targets for osteoporosis treatment. Our preliminary study uses state-of-the-art method, including target-based virtual screening clustering methods to determine promising candidates multitarget properties. Phytochemicals osteoprotective properties from the literature used elucidate molecular interactions toward three targets. The binding scores of compounds normalized rescored. K-means hierarchical applied filter define compounds, silhouette analysis supposed validate method. We explore 108 herbal by cluster approach, find that rutin, sagittatoside A, icariin, kaempferitrin showed strong affinities against integrin. Dockings also provide protein-ligand crucial amino acids binding. provides straightforward less time-consuming approach exploring new further investigations, using combination in silico methods.

Identification of ligands for RNA targets via structure-based virtual screening: HIV-1 TAR

Binding of the Tat protein to TAR RNA is necessary for viral replication of HIV-1. We screened the Available Chemicals Directory (ACD) to identify ligands to bind to a TAR RNA structure using a four-step docking procedure: rigid docking first, followed by three steps of flexible docking using a pseudobrownian Monte Carlo minimization in torsion angle space with progressively more detailed confo...

Nucleotide sequence-based multitarget identification.

MULTIGEN technology (T. Vinayagamoorthy, U.S. patent 6,197,510, March 2001) is a modification of conventional sequencing technology that generates a single electropherogram consisting of short nucleotide sequences from a mixture of known DNA targets. The target sequences may be present on the same or different nucleic acid molecules. For example, when two DNA targets are sequenced, the first an...

3D QSAR pharmacophore based virtual screening and molecular docking for identification of potential HSP90 inhibitors.

Chemical features based 3D pharmacophore models were developed for HSP90 based on the known inhibitors using Discovery Studio V2.1. An optimal pharmacophore model was brought forth and validated using a decoy set, external test set and Fischer's randomization method. The best five features pharmacophore model, Hypo1, includes two hydrogen bond acceptors, three hydrophobic features, which has th...

Structure-Based Virtual Screening Protocol for in Silico Identification of Potential Thyroid Disrupting Chemicals Targeting Transthyretin.

Thyroid disruption by xenobiotics is associated with a broad spectrum of severe adverse outcomes. One possible molecular target of thyroid hormone disrupting chemicals (THDCs) is transthyretin (TTR), a thyroid hormone transporter in vertebrates. To better understand the interactions between TTR and THDCs, we determined the crystallographic structures of human TTR in complex with perfluorooctane...
